ABSTRACT

SUMMARY: The livers of reptiles are being studied as a model for the link between the environment and hepatic tissue. There have been few investigations on the histology of reptile livers, and very few or no studies have examined the histology of liver of veiled chameleon (Chamaeleo calyptratus). This paper describes the histomorphological, histochemical and ultrastructural characterization of the liver of veiled chameleons in southern Saudi Arabia. Seven Chamaeleo calyptratus were captured in the summer season in Abha City, Aseer region, southern Saudi Arabia. Chamaeleon liver samples were processed for histomorphology, histochemistry and ultrastructure analyses. Morphologically liver of Chamaeleo calyptratus was observed as a large dark brown organ with lighter speckles, which represent melanin deposits. It located at the ventral part of abdominal cavity forward of the stomach. Its dimensions approximately were 3.7 x 2 cm. The liver was a bilobed organ divided into two lobes, right and left lobes. The right one was bigger than the others. The gallbladder was well developed and had an elongated shape, situated between the two lobes and contained the bile for the digestion. Microscopically, the liver was found to be covered by a thick layer of connective tissue, which formed the hepatic capsule. Hepatic parenchyma probably appeared in cross sections as hepatic glandular-like alveoli "acini" or follicular structures with various diameters, each acinus contains approximately four to six hepatocytes, surrounded by sinusoidal capillaries filled with abundant melanomacrophages, which are absent in birds and mammals. Melanomacrophages are common in the hepatic parenchyma's perisinusoidal areas, particularly near portal spaces. Hepatocytes are polyhedral or pyramidal with and mostly contained large, rounded nuclei mostly peripherally located, with prominent dark oval nucleoli. Some of nuclei are eccentric or central position. The cytoplasm appeared spongy or vacuolated and more eosinophilic when stained by hematoxylin-eosin and strongly reactive to PAS staining technique, indicating abundant glycogen content. The reticular fibers that surround hepatocytes, blood arteries, and sinusoids supported the hepatic parenchyma. The blood sinusoids are seen interspersed among hepatocytes of varying sizes. The sinusoidal lumen was bordered by flattened endothelial cells and includes elliptical nucleated erythrocytes and liver macrophages as phagocytes, which are also known as Kupffer cells. Branches of the portal vein, hepatic artery, small bile duct, and lymph vessels were detected in the hepatic portal area "tract" or triad which made up of connective. Hematopoietic tissue was observed in subcapsular region and portal triads. Ultrastructurally, the hepatocyte appeared polyhedric containing a single large rounded basal or eccentric vesicular nucleus with prominent nucleolus. Extensive network of rough endoplasmic reticulum (RER) often arranged in an array parallel to the nuclear membrane with many mitochondria, and Golgi apparatus were described. The cytoplasm contained glycogen granules, vesicles or vacuoles scattered throughout the cytoplasm especially at the apical region were reported. The bile canaliculi and the hepatic "Kupffer" cells were also discussed. This is the first study on the histological characterization of the healthy liver of Yemen veiled chameleon in southern Saudi Arabia. The findings reported here should be used as a reference to compare with the pathological abnormalities of the liver in this animal.

ABSTRACT

SUMMARY: Nonalcoholic fatty liver disease (NAFLD) might progress the steatosis to nonalcoholic steatohepatitis (NASH), reaching a cirrhosis state and possibly hepatocellular carcinoma. The liver of three-month-old C57BL/6J mice (wild-type, WT group, n=10) and leptin- deficient obese mice (ob/ob group, n=10) were studied, focusing on the mechanisms associated with the activation of the hepatic stellate cells (HSCs) and pro-fibrogenesis. The obese ob/ob animals' liver showed steatosis, increased lipogenesis gene expressions, inflammation, increased pro-inflammatory gene expressions, inflammatory infiltrate, and potential apoptosis linked to a high Caspase 3 expression. In ob/ob mice, liver sections were labeled in the fibrotic zones by anti-alpha-smooth muscle actin (alpha-SMA) and anti-Reelin, but not in the WT mice. Moreover, the alpha-SMA gene expression was higher in the ob/ob group's liver than the WT group. The pro-fibrogenic gene expressions were parallel to anti- alpha-SMA and anti-Reelin immunofluorescence, suggesting HSCs activation. In the ob/ob animals, there were increased gene expressions involved with lipogenesis (Peroxisome proliferator-activated receptor-gamma, Cell death-inducing DFFA-like effector-c, Sterol regulatory element-binding protein-1c, and Fatty acid synthase), pro-fibrogenesis (Transforming growth factor beta1, Smad proteins- 3, Yes-associated protein-1, Protein platelet-derived growth factor receptor beta), pro-inflammation (Tumor necrosis factor-alpha, and Interleukin-6), and apoptosis (Caspase 3). In conclusion, the results in obese ob/ob animals provide a clue to the events in humans. In a translational view, controlling these targets can help mitigate the hepatic effects of human obesity and NAFLD progression to NASH.


RESUMEN: La enfermedad del hígado graso no alcohólico (HGNA) puede progresar de la esteatosis a esteatohepatitis no alcohólica (ENA), alcanzando un estado de cirrosis y posiblemente carcinoma hepatocelular. Se estudió el hígado de ratones C57BL / 6J de tres meses de edad (tipo salvaje, grupo WT, n = 10) y ratones obesos con deficiencia de leptina (grupo ob/ob, n = 10), centrándose en los mecanismos asociados con la activación de las células estrelladas hepáticas (HSC) y profibrogénesis. El hígado de los animales obesos ob/ob mostró esteatosis, aumento de la expresión génica de la lipogénesis, inflamación, aumento de la expresión génica proinflamatoria, infiltrado inflamatorio y posible apoptosis ligada a una alta expresión de Caspasa 3. En ratones ob/ob, las sec- ciones de hígado se marcaron en las zonas fibróticas con anti-alfa- actina de músculo liso (alfa-SMA) y anti-Reelin, pero no en los ratones WT. Además, la expresión del gen alfa-SMA fue mayor en el hígado del grupo ob/ob que en el grupo WT. Las expresiones génicas profibrogénicas fueron paralelas a la inmunofluorescencia anti-alfa-SMA y anti-Reelin, lo que sugiere la activación de las HSC. ABSTRACT

SUMMARY: Amiodarone (AMD), an orally powerful antidysrhythmic medication that has caused hepatotoxicity on long-term administration, is commonly used across the world. Silymarin ameliorative effects (SLM); this research elucidated the magnitude of the damage to the liver tissue in AMD. We divided 24 albino rats evenly into four groups given daily doses by gastric tube for eight weeks as follows; the 1st group acted as a control group; the 2nd group received SLM; the 3rd group received AMD; and the 4th group received AMD parallel to SLM. Liver tissues prepared for light, electron microscopic and serum samples screened for biomarkers (I)liver injury enzymes, alanine aminotransferase (ALT) and aspartate aminotransferase (AST); (II) oxidative and antioxidant stress, malondialdehyde (MDA) and superoxide dismutase (SOD); and (III) inflammatory markers, tumor necrosis factor-alpha (TNF-a) and interleukin-6 (IL-6). The findings showed that AMD caused hepatic histological changes that included congestion of the blood vessels, leucocytic infiltration and cytoplasmic vacuolation. Ultrastructural degeneration of the mitochondria, endoplasmic reticulum swelling, nuclear pyknosis and increased fat droplets and lysosomes were observed. The biochemical findings showed an increase in the AMD group's ALT and AST activities. The group of rats treated with AMD and SLM, increased the improvements in histology and ultrastructure, while the ALT and AST levels were reduced. Our findings collectively agreed that SLM has a protective impact on AMD hepatotoxicity which can be due to its antioxidant properties.


RESUMEN: La amiodarona (AMD) es un fuerte medicamento antiarrítmico administrado por vía oral que ha causado hepatotoxicidad en la administración a largo plazo utilizado con frecuencia en todo el mundo. Efectos de mejora de la silimarina (SLM); esta investigación analizó la magnitud del daño al tejido hepático en la DMAE. Dividimos 24 ratas albinas de manera uniforme en cuatro grupos que recibieron dosis diarias por sonda gástrica durante ocho semanas de la siguiente manera; el primer grupo fue designado como grupo control; el segundo grupo recibió SLM; el tercer grupo recibió AMD; y el cuarto grupo recibió AMD en paralelo a SLM. Se prepararon tejidos hepáticos para muestras de suero, microscopía de luz y electrónica y se analizaron para biomarcadores (I) enzimas de daño hepático, alanina aminotransferasa (ALT) y aspartato aminotransferasa (AST); (II) estrés oxidativo y antioxidante, malondialdehído (MDA) y superóxido dismutasa (SOD); y (III) marcadores inflamatorios, factor de necrosis tumoral alfa (TNF-a) e interleucina-6 (IL-6). Los hallazgos mostraron que la DMAE genera cambios histológicos hepáticos que incluyen congestión de los vasos sanguíneos, infiltración leucocítica y vacuolación citoplásmica. Se observó una degeneración ultraestructural de las mitocondrias, aumento del retículo endoplásmico, picnosis nuclear y aumento de gotitas de grasa y lisosomas. Los hallazgos bioquímicos mostraron un aumento en las actividades de ALT y AST del grupo AMD. El grupo de ratas tratadas con AMD y SLM, aumentó las mejoras en histología y ultraestructura, mientras que se redujeron los niveles de ALT y AST. Nuestros hallazgos coincidieron colectivamente en que SLM tiene un impacto protector sobre la hepatotoxicidad de AMD debido a sus propiedades antioxidantes.

ABSTRACT

A serous membrane covering the liver and the hepatic parenchyma, consists of hepatocytes, arteries, veins, hepatic sinusoids and biliary ductuli. There are erythrocytes, thrombocytes, melanin particles and Kupffer cell in the hepatic sinusoids and the blood vessels. The gall bladder wall consists of a mucous layer a muscle layer and a serous layer. The bottom of the epithelium abounds with round or oval secretory. In liver, immunohistochemistry results show that AQP1 have intense reaction in hepatic lobule, Kupffer cells (Macrophagocytus stellatus), hepatocytes, portal tract, blood islands, vein and artery, but almost no reaction of AQP2 was detected. In gallbladder, mucous epithelium, endothelial cells from vein and artery all have strong AQP1 expression, AQP2 showed minor diffused positive reaction in gallbladder, which suggesting that AQP1 may have the main role in the absorption and transportation of fluid in hepatobiliary system of Qinghai Lizard.


Una membrana serosa cubre el hígado y el parénquima hepático el cual está formado por hepatocitos, arterias, venas, sinusoides hepáticos y conductos biliares. Se encuentran eritrocitos, trombocitos, partículas de melanina y células de Kupffer en los sinusoides hepáticos y en los vasos sanguíneos. La pared de la vesícula biliar presenta tres capas: mucosa, muscular y serosa. En el hígado, la inmunohistoquímica mostró que AQP1 tiene una reacción intensa en el lóbulo hepático, células de Kupffer, hepatocitos, tracto portal e islotes sanguíneos. En venas y arterias, no se detectó reacción alguna de AQP2. En la vesícula biliar, el epitelio mucoso, las células endoteliales venosas y arteriales tuvieron una importante expresión de AQP1, sin embargo, AQP2 mostró una reacción positiva difusa menor, lo que sugiere que la AQP1 podría tener una función principal en la absorción y transporte de líquido en el sistema hepatobiliar del Lagarto Qinghai.

ABSTRACT

This study aimed to explore the influence of gut microbiota alterations induced by Linderae radix ethanol extract (LREE) on alcoholic liver disease (ALD) in rats and to study the anti-inflammatory effect of LREE on ALD through the lipopolysaccharide (LPS) toll-like receptor 4 (TLR4)-nuclear factor kappa B (NF-κB) pathway. ALD rat models were established by intragastric liquor [50% (v/v) ethanol] administration at 10 mL/kg body weight for 20 days. Rats were divided into six groups: normal group (no treatment), model group (ALD rats), Essentiale group (ALD rats fed with Essentiale, 137 mg/kg), and LREE high/moderate/low dose groups (ALD rats fed with 4, 2, or 1 g LREE/kg). NF-κB and LPS levels were evaluated. Liver pathological changes and intestinal ultrastructure were examined by hematoxylin and eosin staining and transmission electron microscopy. The gut microbiota composition was evaluated by 16S rDNA sequencing. Expression levels of TLR4 and CD68 in liver tissue, and occludin and claudin-1 in intestinal tissue were measured. LREE treatment significantly reduced NF-κB and LPS levels, improved liver pathological changes, and ameliorated intestinal ultrastructure injury. Meanwhile, LREE-fed groups showed a higher abundance of Firmicutes and a lower abundance of Bacteroidetes than the rats in the model group. Administration of LREE suppressed TLR4 overexpression and promoted the expression of occludin and claudin-1 in intestine tissue. Thus, LREE could partly ameliorate microflora dysbiosis, suppress the inflammatory response, and attenuate liver injury in ALD rats. The protective effect of LREE might be related to the LPS-TLR4-NF-κB pathway.

ABSTRACT Objective To investigate the effects of physical training on metabolic and morphological parameters of diabetic rats. Methods Wistar rats were randomized into four groups: sedentary control, trained control, sedentary diabetic and trained diabetic. Diabetes mellitus was induced by Alloxan (35mg/kg) administration for sedentary diabetic and Trained Diabetic Groups. The exercise protocol consisted of swimming with a load of 2.5% of body weight for 60 minutes per day (5 days per week) for the trained control and Trained Diabetic Groups, during 6 weeks. At the end of the experiment, the rats were sacrificed and blood was collected for determinations of serum glucose, insulin, albumin and total protein. Liver samples were extracted for measurements of glycogen, protein, DNA and mitochondrial diameter determination. Results The sedentary diabetic animals presented decreased body weight, blood insulin, and hepatic glycogen, as well as increased glycemia and mitochondrial diameter. The physical training protocol in diabetic animals was efficient to recovery body weight and liver glycogen, and to decrease the hepatic mitochondrial diameter. Conclusion Physical training ameliorated hepatic metabolism and promoted important morphologic adaptations as mitochondrial diameter in liver of the diabetic rats.


RESUMO Objetivo Investigar os efeitos do treinamento físico nos parâmetros morfológicos e metabólicos de ratos diabéticos. Métodos Ratos Wistar foram randomizados para quatro grupos: controle sedentário, controle treinado, diabético sedentário e diabético treinado. Diabetes mellitus foi induzido por administração de Aloxana (35mg/kg) nos Grupos Diabético Sedentário e diabético treinado. O protocolo de treinamento físico incluiu natação com carga de 2,5% do peso corporal, por 60 minutos por dia (5 dias por semana) para os Grupos Controle Treinado e diabético treinado, durante 6 semanas. Ao final do experimento, os ratos foram sacrificados, e o sangue foi coletado para determinação das concentrações séricas de glicose, insulina, albumina e proteínas totais. Amostras do fígado foram coletadas para determinação do glicogênio, proteínas, DNA e diâmetro mitocondrial. Resultados O Grupo Sedentário Diabético apresentou redução no peso corporal, insulinemia e glicogênio hepático, além de maior glicemia e diâmetro mitocondrial hepático. O protocolo de treinamento físico em animais diabéticos foi eficiente para restaurar o peso corporal e o glicogênio hepático, além de reduzir o diâmetro mitocondrial hepático. Conclusão O treinamento físico melhorou o metabolismo hepático e promoveu importantes adaptações morfológicas, como no diâmetro mitocondrial no fígado de animais diabéticos.

ABSTRACT

OBJECTIVES: The number of pancreatic transplants has decreased in recent years. Pancreatic grafts have been underutilized compared to other solid grafts. One cause of discard is the macroscopic appearance of the pancreas, especially the presence of fatty infiltration. The current research is aimed at understanding any graft-related association between fatty tissue infiltration of the pancreas and liver steatosis. METHODS: From August 2013 to August 2014, a prospective cross-sectional clinical study using data from 54 multiple deceased donor organs was performed. RESULTS: Micro- and macroscopic liver steatosis were significantly correlated with the donor body mass index ([BMI]; p=0.029 and p=0.006, respectively). Positive gamma associations between pancreatic and liver macroscopic and microscopic findings (0.98; confidence interval [CI]: 0.95-1 and 0.52; CI 0.04-1, respectively) were observed. Furthermore, comparisons of liver microscopy findings showed significant differences between severe versus absent (p<0.001), severe versus mild (p<0.001), and severe versus moderate classifications (p<0.001). The area under the receiver operating curve was 0.94 for the diagnosis of steatosis by BMI evaluation using a cut-off BMI of 27.5 kg/m2, which yielded 100% sensitivity, 87% specificity, and 100% negative predictive value. CONCLUSIONS: We observed a positive association of macroscopic and microscopic histopathological findings in steatotic livers with adipose infiltration of pancreatic grafts.

ABSTRACT

The liver is one of the major organs that is indirectly affected by cigarette smoke. The aim of this project is to define the histologic and ultrastructural changes in normal liver cells after exposing animals to cigarette smoke. Thirty albino rats were exposed to cigarette smoke for 90 days, followed by morphologic examination of their livers under light microscope and electron microscope. The liver cells of cigarette smoke exposed rats showed mild swelling with increased eosinophilia. Ultrastructural examination of these cells demonstrated cytoplasm with highly proliferated and crowded mitochondria. There were many electron dense mitochondria. These mitochondria were pleomorphic in shape compared to mitochondrias of control rats. Also, loss of mitochondrial cristae and widening of the intermembranous space was noticed. It is concluded that smoking exerts cellular damage and oxidative stress on normal liver cells resulting in ultrastructural changes.


El hígado es uno de los principales órganos indirectamente afectado por el humo del cigarrillo. El objetivo de este trabajo fue definir los cambios histológicos y ultraestructurales de las células normales del hígado después de exponer a los animales al humo del cigarrillo. Treinta ratas albinas fueron expuestas al humo de cigarrillo durante 90 días, seguido de un examen morfológico de los hígados bajo microscopio de luz y microscopio electrónico. Las células hepáticas de las ratas expuestas al humo de cigarrillo mostraron una leve inflamación con un aumento de la eosinofilia. En el examen ultraestructural de estas células se observó el citoplasma mitocondrial altamente proliferado y saturado. Se observó gran cantidad de mitocondrias electrón-densas y éstas presentaban forma pleomórfica en comparación con las mitocondrias del grupo control. Además, se observó pérdida de las crestas mitocondriales y ensanchamiento del espacio intermembranoso. Se concluye que el tabaquismo ejerce daño celular y estrés oxidativo en las células hepáticas normales, lo que resulta en la aparición de cambios ultraestructurales.

AbstractPasteurella multocida is an important veterinary pathogen causing infections in animals and birds. Nowadays, different reports have described the severity of infections, increasing resistance of micro-organisms to antibiotics, and the contribution of ethnoveterinary practices towards the treatment of various ailments of animals. The aim of the present study was to investigate the antibacterial efficacy of the ethanolic extracts of endophytic fungi against P. multocida Capsular Type A strains. A total of six endophytic fungi were isolated from two tropical ethnoveterinary plants: Garcinia xanthochymus H. and Polygonum chinense L. The ethanolic extracts of the endophytic fungi were subjected to in vitro antimicrobial activity by the well diffusion method. Besides, we evaluated the treatment of mice with the potent fungal extract and observed the effects in different organs under electron microscopy. Our results showed that four fungi had antimicrobial activity against the selected pathogen. The best antibacterial activity was showed by the extract of the endophytic fungi, Glomerella magna isolated from G. xanthochymus, with a minimum inhibitory concentration of 46.9 µg/mL and minimum bactericidal concentration of 750 µg/mL. Treatment of mice with the potent fungal extract caused a considerable inhibitory effect on the pathogen growth in vital organs, results that was also confirmed by histopathological studies made by scanning electron microscopy. The present findings indicated that the endophytic fungi G. magna has the potential to provide an effective treatment against infections caused by Pasteurella multocida. ResumenPasteurella multocida es un importante patógeno veterinario que causa infecciones en animales y aves. Hoy en día, diferentes informes han descrito la gravedad de las infecciones, aumentando la resistencia de los microorganismos a los antibióticos, y la contribución de las prácticas etnoveterinarias hacia el tratamiento de diversas enfermedades de los animales. El objetivo del presente estudio fue investigar la eficacia antibacteriana de los extractos etanólicos de hongos endófitos contra cepas de P. multocida tipo capsular A. Un total de seis hongos endófitos fueron aisladas de dos plantas etnoveterinarias tropicales: Garcinia xanthochymus H. y Polygonum chinense L. Los extractos etanólicos de los hongos endófitos fueron sometidos a la actividad antimicrobiana in vitro por el método de difusión. Además, se evaluó el tratamiento de ratones con el extracto de hongos potente y observamos los efectos en diferentes órganos bajo el microscopio electrónico. Nuestros resultados mostraron que cuatro hongos tenían actividad antimicrobiana contra el patógeno seleccionado. La mejor actividad antibacteriana la mostró el extracto de los hongos endófitos, Glomerella magna aislado de G. xanthochymus, con una concentración inhibitoria mínima de 46.9 mg/ml y la concentración bactericida mínima de 750 mg/ml. El tratamiento de ratones con el extracto de hongos potente causó un considerable efecto inhibidor sobre el crecimiento de patógenos en órganos vitales, resultados que también fueron confirmados por estudios histopatológicos realizados por microscopía electrónica de barrido. Los presentes hallazgos indican que el hongos endófitos G. magna tienen el potencial de proporcionar un tratamiento eficaz contra las infecciones causadas por Pasteurella multocida. Sin embargo, el aislamiento de componentes bioactivos necesita más investigación.

ABSTRACT

OBJECTIVE: to identify the perceptions of professionals working in a facility connected with the Brazilian Unified Health System - SUS in regard to what they know, think and talk about public health policy. METHOD: this exploratory-descriptive study with a qualitative nature was conducted with 28 professionals working in a facility connected with the SUS. Data were collected through interviews with guiding questions and analyzed through the thematic content analysis technique. RESULTS: coded and interpreted data resulted in three thematic axes: The SUS - perfect web that does not work in practice; The recurrent habit of complaining about the SUS; The need to rethink the way of thinking about, acting in and managing the SUS. CONCLUSION: the professionals working for the SUS are aware of the principles and guidelines that govern the Brazilian health system, however, they reproduce a dichotomous and linear model of conception and practice strongly linked to the thinking of society in general. .


OBJETIVO: conhecer a percepção de profissionais que atuam em uma instituição conveniada com o Sistema Único de Saúde sobre o que sabem, pensam e falam dessa política pública de saúde. MÉTODO: trata-se de estudo exploratório-descritivo, de caráter qualitativo, realizado com 28 profissionais que atuam em uma instituição conveniada com o Sistema Único de Saúde. Os dados foram coletados por meio de entrevistas com questões norteadoras e analisados pela técnica de análise de conteúdo temática. RESULTADOS: os dados codificados e interpretados resultaram em três eixos temáticos: Sistema Único de Saúde - teia perfeita que não funciona na prática; o recorrente hábito de reclamar do Sistema Único de Saúde; a necessidade de repensar o modo de pensar, atuar e gerir o Sistema Único de Saúde. CONCLUSÃO: os profissionais que atuam no Sistema Único de Saúde têm conhecimento dos princípios e diretrizes que regem o sistema de saúde nacional, no entanto, reproduzem um modelo de concepção e atuação dicotômico, pontual e linear ainda fortemente vigente no pensar da sociedade em geral. .

Los datos fueron recolectados por medio de entrevistas con preguntas orientadoras y analizados con la técnica de análisis de contenido temático. RESULTADOS: los datos codificados y interpretados resultaron en tres ejes temáticos: SUS - red perfecta que no funciona en la práctica; el recurrente hábito de reclamar del SUS; y la necesidad de repensar el modo de pensar, actuar y administrar el SUS. CONCLUSIÓN: los profesionales que actúan en el SUS tienen conocimiento de los principios y directrices que gobiernan el sistema de salud nacional, sin embargo, reproducen un modelo de concepción y actuación dicotómico, puntual, linear y además fuertemente vigente en el pensar de la sociedad en general. .

ABSTRACT

In this work, the morphological features of liver in Chinese Taihe black-bone silky Fowl (BSF) were analyzed by light microscope and transmission electron microscopy. The results showed that two kinds of hepatocytes were present in Taihe BSF liver, i.e., the dark and the light hepatocyte. The dark hepatocyte was electron-dense and lager, with many organelles, mitochondria especially. The light hepatocytes were smaller than the dark. They had electron-lucent cytoplasm with a small number of organelles. Furthermore, there were lipolysosomes in the light hepatocyte. The numerous long and serried finger-like microvilli spread into bile canaliculus lumen. The glycogen granules intensely stained, spread in some hepatocytes. Numerous glycogen granules scattered in cytoplasm especially near bile canaliculi. However, lipid droplets were not observed in any hepatocytes. The natural apoptotic hepatocytes were observed in Taihe BSF liver. The hepatocytes which contain abundant uesicae-like endoplasmic reticulum closed the apoptotic hepatocytes and spread the process to approach the cell residual bodies. Besides, there was a macrophage with several phagosomes. In conclusion, the dark and the light hepatocyte were present in Chinese Taihe BSF liver. They were different from electron-dense and organelles. The hepatocytes of Taihe BSF could undergo natural apoptosis, regeneration and renew ability.


Fueron analizadas las características morfológicas ultraestructurales de hígado en la gallina sedosa china de hueso negro por microscopía óptica y microscopía electrónica de transmisión. Los resultados mostraron que se encontraron hepatocitos claros y oscuros en el hígado de la gallina china Taihe. El hepatocito oscuro era denso y de mayor tamaño. Tenía numerosos organelos, especialmente mitocondrias. Los hepatocitos claros eran más pequeños que los oscuros. El citoplasma presentó un pequeño número de organelos. Además, había lipolisosomas en los hepatocitos claros. Numerosas microvellosidades se extendían hacia los canalículos biliares. En algunos hepatocitos se observó una tinción marcada en los gránulos de glucógeno. Sin embargo, no se observaron gotas de lípidos en los hepatocitos. Se observaron los hepatocitos apoptóticos naturales en el hígado de la gallina Silky Taihe. Aquellos hepatocitos que contenían abundante retículo endoplásmico, cerraban los hepatocitos apoptóticos y extendían el proceso de acercamiento a cuerpos residuales celulares. También hubo un macrófago con varios fagosomas. En conclusión, los hepatocitos claros y oscuros estaban presentes en el hígado de la gallina Taihe china. Estos diferían de electrones de alta densidad y organelos. Los hepatocitos de la gallina Taihe presentaron una apoptosis natural y capacidad de regeneración.

ABSTRACT

The aim of this study was to characterize the normal structure of the liver of Hemisorubim platyrhynchos, a carnivorous freshwater catfish found in Neotropical region, using gross anatomy, histology and histochemistry. Anatomically, the liver presents C-shaped and only two lobes: smaller right and bigger left. The gallbladder is located in right lobe and shows elongated shaped. Histological analysis demonstrated that the hepatic parenchyma is made of two hepatocytes plates surrounded by sinusoids. The hepatocytes are polygonal-shaped cells, with spherical nucleus and a dark prominent nucleolus. The cytoplasm presents large amount of lipids and glycogen deposits PAS positives. There are no hepatic lobules or portal triads. Bile ducts are lined by columnar epithelial cells with apical mucosubstances PAS and AB positive. Furthermore, the liver presents melano-macrophages centers, distributed next to the blood vessels and bile ducts, constituted by cells accumulating pigments, whose presence may be related to the nutritional status of the fish. Moreover pancreatic tissue was observed in visceral portion of liver, formed by exocrine pancreatic tissue and islet organ, constituting an extrahepatic pancreas.


El objetivo de este estudio fue caracterizar la estructura normal del hígado de Hemisorubim platyrhynchos, un bagre carnívoro de agua dulce encontrado en la región neotropical, utilizando anatomía macroscópica, histología e histoquímica de mucosustancias. Anatómicamente, el hígado presenta una forma de C y sólo dos lóbulos de tamaño diferente: menor derecho y mayor izquierdo. La vesícula biliar se encuentra en el lóbulo derecho y presenta forma alargada. El análisis histológico demostró que el parénquima hepático está hecho de dos placas de hepatocitos rodeados por sinusoides. Los hepatocitos son células poligonales con núcleo esférico y un nucléolo oscuro prominente. El citoplasma presenta gran cantidad de lípidos y depósitos de glucógeno PAS positivos. No hay lóbulos hepáticos o triadas portal. Los conductos biliares están revestidos por células columnares epiteliales con mucosustancias apicales PAS y AB positivos. Además, el hígado presenta centros de melanomacrófagos, distribuidos junto a los vasos sanguíneos y conductos biliares, constituidos por células que acumulan pigmentos, cuya presencia puede estar relacionada con el estado nutricional de los peces. Por otra parte, el tejido pancreático se observó en la porción visceral de hígado, formado por tejido pancreático exocrino y órgano islote, que constituye un páncreas extrahepático.

ABSTRACT

The normal liver histology of the African palm squirrel Epixerus ebii was investigated to fill the information gap on its micromorphology from available literature. The liver was covered by a capsule of dense connective tissue- the perivascular fibrous capsule. Beneath this capsule is the liver parenchyma were the hepatocyte were supported by reticular fibres. The hepatocytes in the lobules were hexagonal to polygonal in shape. Some hepatocytes were bi-nucleated. Clear spaces in the parenchyma must be storage sites for lipids in the liver. The classic hepatic lobules presented central vein surrounded by several liver cells. At the portal triad, hepatic vein, hepatic arteries and bile ducts were seen. While the hepatic arteries and veins were lined by endothelium, the bile ducts were lined by simple cuboidal cells. Nerve fibres were also seen in the region of the portal triad. Hepatic sinusoids lined by endothelium were seen in the liver parenchyma between liver lobules. The sinusoids contained macrophages. This report will aid wild life biologists in further investigative research and Veterinarians in diagnosing the hepatic diseases of the African palm squirrel.


Describimos la histología hepática normal de la ardilla de palma africana Epixerus ebii para llenar el vacío sobre su micromorfología en la literatura disponible. El hígado está cubierto por una cápsula de tejido conectivo denso, la cápsula fibrosa perivascular. Debajo de esta cápsula se encuentra el parénquima hepático en el cual fibras reticulares apoyan los hepatocitos. Los hepatocitos en los lóbulos son hexagonales de forma poligonal. Observamos algunos hepatocitos bi-nucleados. Evidentes espacios en el parénquima deben ser emplazamientos de almacenamiento de los lípidos en el hígado. Los lóbulos hepáticos clásicos presentan una vena central rodeada de varias células hepáticas. En la tríada portal, se observó la vena hepática, las arterias hepáticas y los conductos biliares, mientras que las arterias y venas hepáticas estaban revestidas por endotelio, los conductos biliares estaban alineados por células cuboides simples. También se observaron fibras nerviosas en la región de la tríada portal. Sinusoides hepáticos revestidos por endotelio fueron observados entre los lobulillos hepáticos en el parénquima hepático. Los sinusoides contenían macrófagos. El presente estudio ayudará a los biólogos de la fauna silvestre en una investigación más precisa, y a los médicos veterinarios en el diagnóstico de las enfermedades hepáticas en la ardilla de palma africana.

ABSTRACT

Nanoparticles of silver have many important applications and are among the most commonly used nanomaterials. They are increasingly used in a variety of both medical and consumer products which includes: spectrally selective coating for solar energy absorption and intercalation material for electrical batteries, as optical receptors, polarizing filters, catalysts in chemical reaction and bio-labeling. Nanosilver [Ag-NP] has both antibacterial and antiviral activity. Yet, the knowledge about the systemic toxicity of nanosilver is relatively limited. The aim of work: To evaluate the potential toxicity of small size 10nm silver nanoparticles using two different doses [0.1 ml and 0.4 ml] focusing on the ultrastructural changes occurring in mice hepatocytes. This study was performed using three groups of mice. The animals of the first group were given a daily intravenous injection of 0.1 ml of silver nanoparticles for 28 consecutive days. The second group was treated with 0.4 ml of silver nanoparticles for 28 consecutive days. The third group served as a control group in which the animals did not receive any vehicle. The study was focused on the ultrastructure of the liver. Ultrastructure observations of liver cells of mice Treated with any of the two doses [0.1 and 0.4 ml] of 10 nm Ag-NP indicated severe accumulation of dark deposits of Ag-NP in the cytoplasm and the cell organelles. Our study revealed that nanosilver used in doses of 0.1 and 0.4 ml led to deposits in the cells and induced damage of cell components especially the nucleus, mitochondria and chromatin

ABSTRACT

Diabetes mellitus is one of the common and widely distributed metabolic diseases all over the world. This disease is characterized by hyperglycemia that results from defects in insulin secretion, insulin action or both. Different medicinal plant species are used as a traditional treatment for diabetes mellitus e.g. Ambrosia maritima, L. [Damsissa] which is one of these plants that its extract was used to treat diabetic patients long times ago. This work was aimed to investigate the antidiabetic, hypolipidemic and antioxidant effects of the aqueous extract of Ambrosia maritima, L. [Damsissa] on the alloxan-induced diabetic male albino rats. This study was performed on thirty male albino rats with an average 100-110 g body weight. The animals were divided into three groups [10 /cage]; Group I [Control untreated-group], Group II [Alloxan-induced diabetic group] and Group III [diabetic group treated orally with "28.5 mg/ kg body wt. twice/ day" of the plant extract]. The biochemical results showed marked decline [p<0.01] in the levels of the serum insulin, body weight, total proteins, albumin, globulin and HDL accompanied with marked elevation [p<0.001] in the levels of fasting blood glucose, levels of HOMA_IR, AST, ALT, GGT, urea, creatinine, uric acid, serum TC, TG, LDL, VLDL and ratios of TC/HDL and LDL/HDL [risk factors] in diabetic rats in comparison with the control group. Daily management of the diabetic rates with aqueous extract of Damsissa showed significant improvement in most of these parameters. Histologically, considerable improvement in the morphological changes that was observed in diabetic groups had been detected after treatment with Damsissa in liver, kidney and pancreatic tissues in comparison to the control group. It could be concluded that Ambrosia maritima, L. [Damsissa] can be used as an antidiabetic drug that can lower blood glucose concentration and guard against the negative effects of diabetes

Introducción. Ceratopteris pteridoides es un helecho semiacuático de la familia Parkeriacea, ampliamente utilizado en la medicina popular colombiana como diurético y colelitiásico, sobre el cual no existen reportes científicos que avalen su uso popular como diurético. Objetivo. Evaluar el efecto diurético agudo en dosis única y dosis repetidas a corto plazo, de los extractos etanólico y acuoso de C. pteridoides en un modelo in vivo . Materiales y métodos. El extracto etanólico total fue obtenido por maceración de la planta entera de C. pteridoides con etanol y el extracto acuoso fue obtenido por decocción a 60 °C por 15 minutos. Ambos extractos se sometieron a análisis fitoquímico preliminar y estudio histológico posterior a la administración de los extractos durante ocho días consecutivos (1.000 mg/kg). El efecto diurético se evaluó en ratas Wistar, tratadas con los extractos (500 mg/kg), en forma aguda y en dosis repetidas a corto plazo, cuantificando la eliminación de agua y la excreción renal de sodio y potasio por espectrofotometría de absorción atómica y, de cloruros, por titulación mercurimétrica. Resultados. En el modelo agudo, ambos extractos mostraron un significativo efecto diurético y de excreción renal de sodio y potasio en comparación con el control, mientras que con la administración en dosis repetidas a corto plazo mostraron efecto diurético sin eliminación de electrolitos. El estudio histopatológico no sugirió efectos tóxicos hepáticos o renales. Conclusión. Los resultados demuestran la actividad diurética de C. pteridoides y sustentan el uso popular dado a esta planta como diurético en la costa norte colombiana. Se requieren estudios posteriores que permitan aislar e identificar los compuestos responsables de la actividad y los mecanismos de acción involucrados.


Introduction. Ceratopteris pteridoides is a semiaquatic fern of the Parkeriacea family, widely used in the Colombian folk medicine as a diuretic and cholelithiasic, of which there are no scientific reports that validate its popular use. Objective. To evaluate the acute and short-term repeated-dose diuretic effect of the ethanolic and aqueous extracts of C. pteridoides in an in vivo model. Materials and methods. The total ethanolic extract was obtained by maceration of the whole plant of C. pteridoides with ethanol and the aqueous extract by decoction at 60°C for 15 minutes. Both extracts were evaluated in preliminary phytochemical analysis and histological studies after the administration of the extracts for 8 consecutive days (1000 mg/Kg). The diuretic effect was evaluated using Wistar rats treated with the extracts (500 mg/Kg), using an acute and a short-term repeated-dose model, and quantifying water elimination, sodium and potassium excretion by atomic absorption spectrophotometry, and chloride excretion by mercurimetric titration. Results. In the acute model both extracts showed significant diuretic, natriuretic, and kaliuretic effect compared to the control group. Whereas, a short-term repeated-dose administration showed a diuretic effect without elimination of electrolytes. The histopathologic study did not suggest a toxic effect in liver or kidney. Conclusion. The results represent evidence of the diuretic activity of C. pteridoides and give support the popular use given to this plant in the north coast of Colombia. Further studies are required to isolate and identify the compounds responsible for the activity and the mechanism of action involved.

ABSTRACT

Exposure to titanium dioxide nanoparticles [TiO[2] NPs] results from its wide use in the fields of medicine, industry, engineering, and environmental technology. To investigate the effect of administration of TiO[2] NPs on the ultrastructure of the rat liver and renal cortex. Rats were subdivided into two groups: group A [served as control] and group B [TiO[2] group]. TiO[2] was suspended in PBS and administered by an oral gavage to the rats of group B daily for 90 days at a dose of 5 mg/kg body weight. Thereafter, group B was subdivided into group B1 and sacrificed 24 h after the last dose of titanium. Group B2 was left untreated for 12 months and then sacrificed. Specimens from the liver and renal cortex were obtained and processed for examination by transmission electron microscopy. Histopathological changes were detected in the hepatocytes of group B1 in the form of dilated rough endoplasmic reticulum, numerous lysosomes, and abnormal mitochondria. Moreover, accumulation of large lipid droplets and wide cytoplasmic vacuoles was observed. The renal cortex was also affected. Numerous lysosomes were observed in the lining cells of the proximal tubules and the glomerulus showed an apparent increase in the number of mesangial cells. The interstitium was the site of excessive collagen bundles. These hepatic and renal cortical changes were partially ameliorated 12 months after the last dose of TiO[2]. Small doses of TiO[2] NPs for a long duration resulted in a variety of degenerative changes in the rat liver and renal cortex. Therefore, further studies are required to investigate the underlying mechanisms of this toxicity and to search for possible protective measures

ABSTRACT

Corticosteroids have been used extensively since cortisone was first synthesized in the 1950s, but it leads to the development of numerous side effects. Parsley is one of the most effective medicinal herbs. It has antioxidant and free radical scavenging activities. The present study was designed to evaluate the toxic effect of the chronic use of prednisolone on the histological structure of the liver and lung and the protective effect of parsley oil against the changes induced by prednisolone. Twenty-five adult male albino rats were used. They were divided into three groups: group I was the control group; group II received 2 mg/kg/day prednisolone once daily for 1 month; and group III received parsley oil 0.6 ml/kg/day once daily followed by prednisolone after 2 h for 1 month. The liver and the lung were dissected out and processed for light and electron microscopic examination. In addition, a statistical study was carried out for determining the thickness of the interalveolar septum. In group II, some hepatocytes showed vacuolations and fatty changes and other hepatocytes revealed apoptotic changes. Lung tissue revealed sloughing of the bronchiolar epithelium. Hemorrhage and cellular infiltration were seen in airway spaces. Moreover, there was highly significant thickening of the interalveolar septa. Apparent increase in atypical type II pneumocytes with hyperchromatic nuclei was seen. Electron microscopy revealed vacuolated hyperchromatic type II pneumocytes. In group III, liver cells were normal except for some vacuolations in the cytoplasm. The lung continued to reveal significant increase in thickness of the interalveolar septa. Some type II pneumocytes showed hyperchromatic features. Electron microscopy revealed type II pneumocytes with a few lamellar bodies. Compared with the liver, the dose of parsley and its duration of treatment with respect to the lung should be markedly higher to have a significant effect, as the destructive effect of prednisolone on the lung is greater than that on the liver

ABSTRACT

Sildenafil citrate [SC] is an effective drug for treatment for erectile dysfunction, which is a common symptom in patients with liver disease. The effect of SC on liver structure is elusive and has not been thoroughly studied. The aim of this study was to investigate the effect of SC treatment for different periods and its withdrawal on the liver structure in adult male rats. Twenty-four adult male albino rats were classified into four groups: group I [control], group II [rats treated with SC orally at a dose of 10 mg/kg/day for 4 weeks], group III [rats treated with 10 mg/kg/day SC for 8 weeks] and group IV [recovery group; rats treated similarly as group III and then left for 4 weeks without SC treatment]. Liver specimens were processed for histological, histochemical and immunohistochemical study. In group II, the liver revealed significant enlargement of many hepatocytes with clumping of cell organelles in their cytoplasm. There were frequent Kupffer cells, activated hepatic stellate cells and increased deposition of collagen fibers. In group III, these changes were more marked, in addition to reduced glycogen staining in hepatocytes. Dilated bile ductules and cellular infiltration were noticed in many portal tract areas. In the recovery group, most changes were relatively reduced but not reversed completely. SC exerted deleterious structural effects on the hepatic tissue that were more pronounced with longer duration of SC administration. Some of these deleterious effects were reversible after discontinuation of the drug

ABSTRACT

Hepatic fibrogenesis is a common result of liver injury. It is believed to be a critical factor that leads to hepatic failure. A critical event in fibrogenesis is activation of hepatic stelate cells [HSCs]. The aim of this investigation was to study the role of HSCs in figrogenesis in a model of pomegranate juice [PJ]-treated fatty liver induced by junk food using immunohistochemical and electron microscopic study. Thirty young male albino rats were divided into control [1] and experimental [II] groups. Group II was further divided into two subgroups: II-a [junk food] and II-b [pomegranate juice + junk food]. After 8 weeks, blood samples were collected for detection of leptin and tumour necrosis factor alpha TNF-alpha. Then half of the liver samples were processed for light microscopic examination, whereas the other half were prepared for electron microscopic examination. Praffin sections were stained using H and E, glial ibrillary acid protein, alpha -smooth muscle actin, TNF- alpha, and transforming growth factor -beta-1 TGF-beta1. Morphometric and statistical studies for assessing immunoexpression were carried out. HSC's markers glial fibrilar acid protein and alpha -smooth muscle actin and cytokines TNF- alpha and TGF- beta1 in subgroup II-a showed strong positive immunoexpression. Electron microscopic study showed activated - HSCs containing granules and collagen fibrils. Proliferative and myofibroblast -HSCs were also seen in the same group. Subgroup II-b showed a nonsignificant increase in immunoexpression of HSC s markers and cytokines. However, only activated -HSCs were seen. Immunoexpression of HSC markers and cytokines may be used as an indicator for liver fibrosis. Presence of different types of HSCs in fatty liver explains their role in fibrosis. Further experimental and clinical studies directed toward inhibiting the activity of HSC may delay or prevent liver fibrosis occurs in many pathological conditions
